Ginkgo’s PichiaWorks Protein Expression Kit is a Pichia pastoris based system designed for  high-level production of cytosolic and secreted recombinant proteins. The kit provides sufficient material to build five Pichia strains. 

PichiaWorks builds on all of the advantages of P. pastoris, but notably does not use methanol for induction. Instead, it has been engineered for high expression with just a simple glucose-induction protocol.  The kit contains all required items for building highly expressing Pichia strains: competent cells with landing pads for your gene of interest and the molecular machinery that enables high-efficiency integration.

PichiaWorks Protein Expression System

We evaluated mCherry expression in Pichia under three promoters over a 72-hour flask fermentation: pGAP (constitutive), pAOX1 (methanol-inducible), and pSynPro (proprietary PichiaWorks strain specific). All strains were genetically engineered with a single-copy expression cassette at the same chromosomal locus.

The PichiaWorks strain significantly outperformed traditional systems, showing higher fluorescence by 48 hours and achieving maximum intensity at 72 hours. Unlike the labor-intensive methanol induction, which requires biomass pelleting and resuspension, the PichiaWorks system simplifies the process, achieving higher protein yields by a simple 2% v/v preculture inoculation into the production media.
