We evaluated mCherry expression in Pichia under three promoters over a 72-hour flask fermentation: pGAP (constitutive), pAOX1 (methanol-inducible), and pSynPro (proprietary PichiaWorks strain specific). All strains were genetically engineered with a single-copy expression cassette at the same chromosomal locus.
The PichiaWorks strain significantly outperformed traditional systems, showing higher fluorescence by 48 hours and achieving maximum intensity at 72 hours. Unlike the labor-intensive methanol induction, which requires biomass pelleting and resuspension, the PichiaWorks system simplifies the process, achieving higher protein yields by a simple 2% v/v preculture inoculation into the production media.
