The DSB Booster (20x) is an optimized formula designed to enable the formation of disulfide bonds during cell-free protein synthesis. It establishes the redox environment your target protein needs to pair its cysteines correctly, so that more of what you synthesize comes out of the reaction correctly folded and functional.

This 20x concentrated solution is included as a part of Ginkgo's CFPS Disulfide Premium kit, and can be purchased separately for users who want to titrate beyond the amount supplied in the kit.

Technical Information:

  • The standard dose is 1 µL per 20 µL reaction (1x, 5% v/v).
  • Available sizes are 50 μL, which supports 50 reactions at 1x, and 500 μL, which supports 500 reactions at 1x.
  • Thaw on ice before use and minimize freeze-thaw cycles.
  • Store at −80°C.
  • The product ships on dry ice.

DSB Booster - Beta release

DSB Booster dose response scales with disulfide-bond complexity. Relative yield of three model targets spanning a range of disulfide-bond content (VHH, 1 to 2 disulfides; Gaussia luciferase, 5 disulfides; reteplase, 9 disulfides) as a function of DSB Booster dose. Values are normalized within each target to the yield obtained at the standard 1x booster dose (solid bars). Hatched segments represent the incremental yield delivered by increasing booster dose above 1x, with labels giving the percentage gain relative to 1x. Error bars show standard deviation of replicate reactions (n = 3)
